以印度南瓜种(Cucubita maxima Duch.)“北观”自交系为试材,研究“北观”南瓜子叶节高效快速离体再生体系的构建。结果表明,剥壳种子用0.1%Hg Cl2溶液消毒9 min,无菌催芽后选取子叶微展、绿色的苗态制作子叶节,接种到添加1.0 mg/L 6-BA的MS培养基上诱导不定芽,诱导率达64.5%,子叶节平均再生不定芽3.06个。生长健壮的不定芽转接到MS+NAA 1.0 mg/L+活性炭2.0 g/L培养基上生根培养,主根达2~3条时驯化移栽,发育成完整植株。
The purpose of this study was to evaluate the efficient and rapid in vitro regeneration system of pumpkin cotyledonary node in Cucubita maxima Duch. The results showed that the peeled seeds were disinfected with 0.1% Hg Cl2 for 9 min. After sterile germination, the cotyledons were selected for microscopic growth. The green seedlings were made into cotyledonary nodes and inoculated on MS medium supplemented with 1.0 mg / L 6-BA. Bud, the induction rate was 64.5%, and the cotyledonary node regenerated an average of 3.06 adventitious buds. Growing adventitious buds were transplanted into MS + NAA 1.0 mg / L + activated carbon 2.0 g / L medium for rooting, with 2 to 3 main roots transplanted and grown into complete plants.
