枸杞子经热水提取,用蛋白酶法和Sevag法除蛋白,透析纯化得到枸杞多糖(Ivciumbarbarumpolysaccharide,LBP).经UV扫描,LBP未见核酸(260nm)和蛋白质(280nm)特征吸收峰,PAGE电泳和Bradford法测蛋白质含量,仅测到微量蛋白质及低于5mg/g糖,红外分析显示有典型多糖吸收峰,对其活性作了分析,结果表明本室所提取的LBP具有明显增强ConA和LPS诱导的小鼠脾脏淋巴细胞增殖作用,并对S-180荷瘤小鼠具明显的免疫抑瘤作用。
Gardenia jasminoides were extracted by hot water, protein was removed by protease method and Sevag method, purified by dialysis and purified to obtain IBP (Ibium barbarum polysaccharide) (LBP). After UV scanning, the characteristic absorption peaks of nucleic acid (260 nm) and protein (280 nm) were not detected in LBP. Protein content was measured by PAGE and Bradford method. Only trace amounts of protein and less than 5 mg/g sugar were detected. Infrared analysis showed typical polysaccharide absorption peaks. The activity was analyzed and the results showed that the LBP extracted from this chamber had significantly enhanced ConA and LPS-induced splenic lymphocyte proliferation in mice and had a significant immunosuppressive effect on S-180 tumor-bearing mice.
