目的本研究应用GK糖尿病大鼠心肌梗死模型观察血管内皮生长因子(VEGF)在慢性氯化钴处理后GK糖尿病大鼠心肌梗死后的表达。方法应用体质量在200~250g的雄性GK糖尿病大鼠及其对照雄性Wistar大鼠,通过开胸手术结扎麻醉大鼠冠状动脉左前降支制作大鼠在体心肌梗死模型。心肌梗死模型制作前测量血糖。手术成功后分组饲养大鼠3周。3周后,测量血糖,活体剪取心脏,测量心肌梗死面积,进行VEGF的免疫组织化学染色和VEGF反转录聚合酶链反应(RT-PCR)检测。结果①慢性氯化钴处理降低GK糖尿病大鼠血糖及心肌梗死面积,而对Wistar大鼠血糖及心肌梗死面积无影响;②慢性氯化钴处理增加VEGF在GK糖尿病大鼠缺血区的表达,而对Wistar大鼠VEGF表达没有影响。结论 VEGF在慢性氯化钴处理后GK糖尿病大鼠缺血区的表达增加,降低GK糖尿病大鼠心肌梗死面积。
Objective To investigate the expression of vascular endothelial growth factor (VEGF) after myocardial infarction in GK diabetic rats treated with chronic cobalt chloride by using GK diabetic rat myocardial infarction model. Methods A rat model of myocardial infarction was established by ligating left anterior descending coronary artery of anesthetized rats with male GK diabetic rats of 200-250 g body weight and its control male Wistar rats. Myocardial infarction model before the measurement of blood glucose. Rats were housed for 3 weeks after successful operation. After 3 weeks, the blood glucose was measured, the heart was excised from the living body, the area of myocardial infarction was measured, the immunohistochemical staining of VEGF and VEGF reverse transcription polymerase chain reaction (RT-PCR) were performed. Results ① Chronic treatment of cobalt chloride decreased blood glucose and myocardial infarction area in GK diabetic rats, but had no effect on the area of blood glucose and myocardial infarction in Wistar rats. ② Chronic cobaltous chloride treatment increased the expression of VEGF in ischemic area of GK diabetic rats, But had no effect on VEGF expression in Wistar rats. Conclusion The expression of VEGF in the ischemic area of GK diabetic rats increases after chronic cobalt chloride treatment, and decreases the area of myocardial infarction in GK diabetic rats.
