目的研究当归水提物多成分体内代谢动态变化过程,筛选当归的质控成分,寻找中药质量评价新方法。方法采用肠系膜静脉和腹主动脉取血的方法,运用HPLC对比分析当归经肠代谢和肝代谢的化学成分特征谱,推断当归水提物中多成分的代谢变化过程。结果分析的15种成分在消化液中均基本稳定,其中有4种成分可能被肠道代谢,7种成分被肝脏代谢。并发现1种新成分是肝代谢产物,7种新成分是肠代谢产物,但其中5种肠代谢产物经肝脏又被代谢转化,未能吸收入血。最终确定了当归水提物中的4种原型成分和3种肠肝代谢产物能吸收入血。结论综合分析当归各成分的代谢轮廓,初步筛选出包括洋川芎内酯I在内的4种原型成分作为质控成分,建立了“质代关联”的中药质量评价方法。
OBJECTIVE To study the dynamic process of metabolism of the constituents of Angelica sinensis aqueous extract and to screen the quality control components of Angelica sinensis to find out a new method for evaluating the quality of traditional Chinese medicine. Methods The mesenteric vein and the abdominal aorta blood were collected. The chemical constituents of Angelica sinensis, gut metabolism and hepatic metabolism were analyzed by HPLC to deduce the metabolic process of the constituents. Results Analysis of the 15 components in the digestive juice were basically stable, of which four kinds of ingredients may be intestinal metabolism, seven kinds of components are metabolized by the liver. And found that a new component is liver metabolites, seven kinds of new ingredients are intestinal metabolites, but five of which intestinal metabolic products by the liver and metabolic transformation, failed to absorb into the blood. Finally, four kinds of archetypal components and three kinds of intestinal metabolites of Angelica sinensis water extract can be absorbed into the blood. Conclusion The metabolic profiles of each component of Angelica sinensis were comprehensively analyzed. Four prototype components including yangchongxiong I were initially screened out as the quality control components, and the quality evaluation method of “quality control” was established.
