目的研究人牙周膜干细胞与牙周膜非干细胞之间差异表达的microRNA(miRNA)的靶基因并对其功能进行预测分析。方法本研究于2011年7月至2013年10月在福建医科大学附属口腔医院进行。采用基因芯片技术筛选人牙周膜干细胞与牙周膜非干细胞之间差异性表达的miRNA,并通过生物信息学对差异表达的miRNA进行靶基因预测及靶基因功能富集分析。结果与人牙周膜非干细胞相比,人牙周膜干细胞中明显上调的关键miRNA 25个,调控的关键靶基因21个;明显下调的关键miRNA 5个,调控的关键靶基因9个。结论特异性表达的miRNA主要调控的靶基因及其基因功能与人牙周膜干细胞的干性维持密切相关。
Objective To study the target gene of differentially expressed microRNA (miRNA) between human periodontal ligament stem cells and periodontal ligament non-stem cells, and to predict its function. Methods The study was performed at the Affiliated Stomatological Hospital of Fujian Medical University from July 2011 to October 2013. Gene microarray was used to screen miRNAs differentially expressed between human periodontal ligament stem cells and periodontal ligament non-stem cells. Bioinformatics methods were used to predict differentially expressed miRNAs and enrichment of target genes. Results Compared with human periodontal ligament non-stem cells, there were 25 key miRNAs that were significantly up-regulated in human periodontal ligament stem cells and 21 key target genes were regulated, 5 of which were significantly down-regulated and 9 of which were key genes of regulation. Conclusion The target gene and its gene function regulated by miRNAs specifically expressed are closely related to the dry maintenance of human periodontal ligament stem cells.
