以种子特异性表达的大豆凝集素基因启动子、终止子以及棉株体内硬脂酰 -ACP△ 9脱氢酶的基因 gh SAD-1进行倒位重复基因构建 ,以农杆菌介导法转化陆地棉品种 Coker3 1 5,获得了较高的转化频率。PCR、Southern杂交及种子内脂肪酸组分含量分析。结果表明 ,该倒位重复基因的表达及沉默效应在不同的转基因品系间存在差异 ,硬脂酸含量分别由对照的 2 %提高至 4%~3 8.2 % ,而棕榈酸含量则明显降低。高含硬脂酸的转基因品系可作为棉子油品质改良的基础材料 ,具有极大的潜在利用价值
The gene of soybean lectin gene, the terminator, and the gh SAD-1 gene of stearoyl-ACP △ 9 dehydrogenase in cotton were inoculated by seed-specific inversions. The Agrobacterium-mediated transformation of terrestrial Cotton variety Coker3 1 5, obtained a higher conversion frequency. PCR, Southern hybridization and content analysis of fatty acids in seeds. The results showed that the expression and silencing effect of the inverted repeat gene was different in different transgenic lines. The content of stearic acid increased from 2% to 4% ~32.2% of control, while the content of palmitic acid decreased obviously. High-stearic acid-containing transgenic lines can be used as the basic material for improving the quality of cottonseed oil, with great potential value
