目的观察生殖道沙眼衣原体(CT)初次感染后妊娠大鼠在胚胎着床窗口期子宫蜕膜C3a,γ干扰素(IFN-γ)和白血病抑制因子(LIF)表达量的变化。方法选择成年健康状况良好的雌性SD大鼠35只(除去不符合标准的雌鼠5只,最终以30只作统计学分析),随机均分为实验组和对照组两组。实验组通过阴道接种沙眼衣原体D型株,对照组阴道注射等量生理盐水。分别在大鼠妊娠后第5、第6和第7天(即胚胎着床窗口期)处死大鼠,并从子宫取材,通过免疫组织化学法定量检测在胚胎着床窗口期子宫蜕膜C3a、IFN-γ和LIF表达量。结果 30只大鼠子宫蜕膜组织均有C3a、IFN-γ和LIF表达。实验组C3a和IFN-γ表达量均低于对照组(P<0.01);实验组在妊娠第5天和第6天LIF比对照组低表达(P<0.01),妊娠第7天两组表达量无显著差异(P>0.05)。实验组中IFN-γ和C3a在子宫蜕膜中的表达呈正相关(r=0.9965),IFN-γ与LIF的表达无相关性(r=0.2331)。结论生殖道感染CT后,子宫局部C3a、IFN-γ和LIF表达量有变化并与着床有密切联系。
Objective To observe the changes of the expression of C3a, IFN-γ and leukemia inhibitory factor (LIF) in uterine decidua of pregnant rats after primary infection of genital tract Chlamydia trachomatis (CT). Methods Thirty-five adult female Sprague-Dawley rats were randomly divided into experimental group and control group, with 35 females out of standard and 30 females excluded. The experimental group was inoculated intraperitoneally with D type of Chlamydia trachomatis by vaginal instillation, and the control group was injected with the same amount of normal saline vaginally. The rats were sacrificed on the 5th, 6th and 7th day after embryo implantation (ie, the embryo implantation window), and then taken from the uterus. Immunohistochemistry was used to detect the expression of C3a, IFN-γ and LIF expression levels. Results The expression of C3a, IFN-γ and LIF in uterine deciduas of 30 rats were detected. The expression of C3a and IFN-γ in the experimental group were lower than those in the control group (P <0.01). The expression of LIF in the experimental group was lower than that in the control group on the 5th and 6th day of pregnancy (P <0.01) There was no significant difference (P> 0.05). There was a positive correlation between the expression of IFN-γ and C3a in the decidua (r = 0.9965) and the expression of IFN-γ and LIF in the experimental group (r = 0.2331). Conclusion After CT of genital tract infection, the expression of C3a, IFN-γ and LIF in uterus varies with the close relationship with implantation.
