目的:探讨不同浓度桂皮醛液(cinnamaldehyde,CA)对成骨细胞成骨功能及BMP-2表达的影响。方法:以0(对照组)、10、20、40、80、160、320μg/mL的CA作用于小鼠颅顶前骨细胞亚克隆14(MC3T3-E1 subclone 14,MC3T3),MTT法检测在CA作用1、3、5 d后的增殖情况,分别在3、5、7 d检测碱性磷酸酶(alkaline phosphatase,ALP)活性,以观察6种浓度的CA对成骨细胞成骨功能的影响;荧光实时定量PCR(quantitative real-time PCR,RT-q PCR)检测BMP-2 mRNA在低浓度组(10、20μg/ml)CA作用1、3、5 d后的表达。采用SPSS 17.0软件包对数据进行统计学分析。结果:MTT显示,10、20μg/mL的CA作用于MC3T3后,与空白对照组相比,吸光度值稍有下降但差异不显著(P>0.05),ALP活性显著提高;RT-qPCR结果显示,10、20μg/mL的CA促进BMP-2 mRNA的表达(P<0.05),40、80、160、320μg/mL的CA可抑制成骨细胞增殖、ALP活性及BMP-2 mRNA的表达(P<0.05),随着浓度升高,抑制作用更明显。结论:低浓度CA可提高成骨细胞的ALP活性,促进成骨和BMP-2 mRNA表达,其机制可能是通过上调BMP-2基因表达。
Objective: To investigate the effects of different concentrations of cinnamaldehyde (CA) on osteoblast osteoblast function and BMP-2 expression. Results: MTT showed that after treated with 10, 20μg / mL CA for 3 days, the absorbance value of MC3T3 slightly decreased but the difference was not significant (P> 0.05), while the activity of ALP was significantly increased. The results of RT- CA at 10 and 20μg / mL could promote the expression of BMP-2 mRNA (P <0.05). CA at 40,80,160 and 320μg / mL could inhibit the proliferation of osteoblasts, the activity of ALP and the expression of BMP- 0.05), with the concentration increased, the inhibitory effect is more obvious. CONCLUSION: Low concentrations of CA can increase the ALP activity of osteoblasts and promote the osteogenesis and BMP-2 mRNA expression. The mechanism may be through up-regulating BMP-2 gene expression.
