目的探讨miR-106b-25基因簇(miR-93,miR-106b)在肝泡型包虫病中的表达及靶基因预测。方法Balb/c小鼠40只随机分为实验组24只(注射0.1 ml泡球蚴组织匀浆液)、对照组15只(注射0.1 ml生理盐水),正常组1只(不作任何处理)。分别收集实验组和对照组小鼠1月、3月和6月肝脏标本,提取RNA并定量测定miR-93和miR-106b的相对表达量,预测miR-93和miR-106b的靶基因。结果在感染3月实验组小鼠的肝内miR-93和miR-106b相对表达量分别为3.704±1.776和5.901±2.921,明显高于对照组和正常组,差异具有统计学意义(P<0.05);实验组1月和6月的相对表达量与对照组和正常组比较,差异均无统计学意义(P>0.05);经生物信息数据库比对后可知p21可能是miR-93、miR-106b共同的目的基因,主要发挥调控细胞周期及参与细胞凋亡等功能。结论miR-93和miR-106b在肝泡型包虫病中的表达差异可能与靶基因p21有关。miR-106b-25基因簇可能参与泡球蚴肝损伤的过程。
Objective To investigate the expression of miR-106b-25 gene cluster (miR-93, miR-106b) in hepatic hydatid cyst and predict the target gene. Methods Forty Balb / c mice were randomly divided into experimental group (n = 24), control group (injected 0.1 ml normal saline) and normal group (n = 24). The liver samples from January, March and June in experimental group and control group were collected and RNA was extracted and the relative expression levels of miR-93 and miR-106b were quantified. The target genes of miR-93 and miR-106b were predicted. Results The relative expression levels of miR-93 and miR-106b in the liver of experimental mice infected with 3 months were 3.704 ± 1.776 and 5.901 ± 2.921, respectively, which were significantly higher than that of the control group and the normal group (P <0.05 ). Compared with the control group and the normal group, there was no significant difference in the relative expression level of the experimental group between January and June (P> 0.05); compared with the biological information database, p21 may be the expression of miR-93, miR- 106b common target gene, mainly play a role in regulating the cell cycle and participate in apoptosis. Conclusion The difference of expression of miR-93 and miR-106b in hepatic echinococcosis may be related to the target gene p21. The miR-106b-25 gene cluster may be involved in the process of AEC in liver injury.
