目的:观察中风皂贝化痰胶囊对大鼠脑缺血再灌注损伤后神经保护作用及血管内皮生长因子(vascularendothelial growth factor,VEGF)表达的影响。方法:50只SD大鼠随即分成:假手术组,缺血再灌注组,脑缺血再灌注+中风皂贝化痰胶囊高、中、低剂量组(1.62,1.08,0.54 g.kg-1.d-1)。采用Zea-Longa方法制备大鼠脑缺血再灌注模型,观察中风皂贝化痰胶囊对大鼠脑缺血再灌注损伤后的神经保护作用及VEGF表达的影响。结果:与假手术组相比,缺血再灌注大鼠出现严重的神经功能缺失症状,皮质和海马区VEGF表达增强;与脑缺血再灌注组相比,中风皂贝化痰胶囊治疗组能显著改善大鼠神经功能缺失症状,增加VEGF的表达,其中以中风皂贝化痰胶囊高剂量组的神经保护作用最为显著,VEGF阳性细胞区积分吸光度为(150.8±9.8)。结论:中风皂贝化痰胶囊对大鼠脑缺血再灌注损伤具有保护作用,其作用机制可能与增加VEGF的表达有关。
Objective: To observe the effect of SSP on the neuroprotection and the expression of vascular endothelial growth factor (VEGF) after cerebral ischemia-reperfusion injury in rats. Methods: Fifty SD rats were randomly divided into sham operation group, ischemia-reperfusion group, cerebral ischemia-reperfusion + stroke SBBT high, middle and low dose groups (1.62,1.08,0.54 g.kg-1 .d-1). The model of cerebral ischemia-reperfusion was established by Zea-Longa method. The neuroprotective effect of SSP and the expression of VEGF were observed after cerebral ischemia-reperfusion injury in rats. Results: Compared with the sham-operated group, the symptoms of severe neurological deficit in ischemia-reperfusion rats were increased, and the expression of VEGF in cortex and hippocampus was enhanced. Compared with the cerebral ischemia- reperfusion group, the stroke- Significantly improved the neurological deficits in rats and increased the expression of VEGF. The neuroprotective effect was highest in the high-dose Siberian Saponin Capsule group, and the integral absorbance of VEGF-positive cells was (150.8 ± 9.8). Conclusion: Siberian Saponification Capsule has a protective effect on cerebral ischemia-reperfusion injury in rats and its mechanism may be related to the increase of VEGF expression.
