应用蛋白质芯片技术筛选早中期和晚期肺癌患者血清差异蛋白

目的 应用蛋白质芯片技术检测早中期和晚期肺癌患者血清中的差异表达蛋白并探讨其临床意义.方法 应用表面增强激光解析电离飞行时间质谱(SELDI-TOF-MS)技术,使用弱阳离子交换(WCX-2)蛋白芯片分别检测两组肺癌患者血清蛋白指纹图谱,采用蛋白质芯片阅读机读取数据,CiphergenProtein Chip 3.0版本的分析软件采集数据,读取峰信息,进行统计学处理.两组之间蛋白质峰强度的比较采用X2检验.结果 早中期和晚期肺癌患者血清中存在10个有统计学意义的差异蛋白质峰.与早中期相比,晚期肺癌患者血清中高表达的蛋白质相对分子质量为7978、8139、15 951和16 133,晚期肺癌患者血清中低表达的蛋白质相对分子质量为2867、6885、8701、8840、13 781、13 955.结论 SELDI-TOF-MS蛋白质芯片技术是一种快速、简单、敏感、样本用量少和高通量的蛋白质检查分析方法,可以直接筛选早中期和晚期肺癌患者血清中的差异蛋白,可能成为远处转移的诊断指标,具有较好的临床应用价值.
