目的:研究细胞周期蛋白D1(cyclin D1)siRNA对口腔鳞癌细胞Tca8113凋亡的作用。方法:采用cyclin D1-siRNA慢病毒感染Tca8113,qPCR检测cyclin D1表达,MTT检测细胞活性,Annexin-V单染检测细胞凋亡。结果:Cyclin D1-siRNA慢病毒成功抑制Tca8113中cyclin D1mRNA表达。Cyclin D1干扰组细胞活性显著低于阴性病毒组(P<0.01),且细胞凋亡率高于阴性对照组(P<0.01)。结论:Cyclin D1干扰能促进Tca8113凋亡,起到抗肿瘤效应。
Objective: To study the effect of cyclin D1 siRNA on the apoptosis of oral squamous cell carcinoma cell line Tca8113. Methods: The cyclin D1-siRNA lentivirus was used to infect Tca8113. The expression of cyclin D1 was detected by qPCR. The cell viability was detected by MTT and the apoptosis was detected by Annexin-V staining. Results: Cyclin D1-siRNA lentivirus successfully inhibited cyclin D1 mRNA expression in Tca8113. The cell viability of Cyclin D1 group was significantly lower than that of negative group (P <0.01), and the apoptosis rate of Cyclin D1 group was higher than that of negative group (P <0.01). Conclusion: Cyclin D1 interference can promote Tca8113 apoptosis and play anti-tumor effect.
