目的探讨C-RP、APN在IR大鼠各组织的表达情况。方法19只SD大鼠,随机分为对照组(NC,n=9)与高脂组(HF,n=10),饲养20周后,行OGTT和胰岛素释放试验(IRT),采用实时荧光定量PCR(RT-PCR)及Northern blot法检测大鼠肝脏、脂肪组织C-RP及APN的表达。结果与NC组比较,HF组血糖、胰岛素水平增加,表明已处于IR状态。Northern blot检测显示,HF组脂肪组织中APN的表达较NC组下降(P<0.01)。RT-PCR显示,HF组肝脏组织C-RP较NC组上升约62%(P<0.01),脂肪组织中C-RP的表达较NC组上升约88%(P<0.01)。结论高脂饲养诱导的IR大鼠,肝脏、脂肪组织C-RP表达增加,而脂肪组织APN表达下降。
Objective To investigate the expression of C-RP and APN in various tissues of IR rats. Methods Twenty-nine SD rats were randomly divided into control group (NC, n = 9) and hyperlipidemia group (HF, n = 10). After 20 weeks of feeding, OGTT and insulin release test (IRT) The expression of C-RP and APN in rat liver and adipose tissue were detected by PCR (RT-PCR) and Northern blot. Results Compared with NC group, the level of blood glucose and insulin in HF group increased, indicating that it was in IR state. Northern blot showed that the expression of APN in adipose tissue of HF group was lower than that of NC group (P <0.01). RT-PCR showed that C-RP in liver increased about 62% (P <0.01) in HF group and C-RP increased about 88% (P <0.01) in adipose tissue compared with NC group. Conclusion The expression of C-RP in liver, adipose tissue and the expression of APN in adipose tissue were decreased in high-fat diet-induced IR rats.
