目的研究特发性血小板减少性紫癜(ITP)患者血浆中可溶性CD40配体(sCD40L)及T、B淋巴细胞和血小板上CD40L的变化,探讨CD40分子在ITP中的作用。方法应用酶联免疫吸附法检测患者血浆中sCD40L的变化;流式细胞术检测不同细胞上CD40L的表达水平。结果 ITP患者血浆中的sCD40L浓度明显高于正常对照组(P<0.05),且血小板计数对血浆中sCD40L浓度有一定程度的影响(r=0.58,P<0.05);ITP患者T淋巴细胞上CD40L的表达率较正常对照组明显升高(P<0.05)。结论 ITP患者血浆及T细胞上CD40L升高,淋巴细胞表面的CD40-CD40L相互作用可能与ITP的发病有关。
Objective To study the changes of plasma soluble CD40 ligand (sCD40L), T, B lymphocytes and platelet CD40L in idiopathic thrombocytopenic purpura (ITP) patients and to explore the role of CD40 in ITP. Methods The changes of sCD40L in plasma were detected by enzyme-linked immunosorbent assay. The expression of CD40L on different cells was detected by flow cytometry. Results The concentration of sCD40L in plasma of ITP patients was significantly higher than that of normal controls (P <0.05), and the platelet count had a certain effect on the concentration of sCD40L in plasma (r = 0.58, P <0.05) The expression rate was significantly higher than the normal control group (P <0.05). Conclusions The level of CD40L on plasma and T cells in patients with ITP is elevated, and the interaction of CD40-CD40L on lymphocytes may be related to the pathogenesis of ITP.
