目的 建立一种简便可靠的脑微血管内皮细胞的培养方法。方法 新生SD乳鼠脑皮质经匀浆、过滤、消化和差速贴壁等方法对大鼠脑微血管内皮细胞进行原代培养及传代培养 ,对培养的细胞进行形态学观察 ,并用Ⅷ因子相关抗原进行免疫细胞化学鉴定。结果 培养的细胞呈单层贴壁生长 ,约 7~ 9d后呈典型的“铺路石”征象 ,免疫细胞化学鉴定证实为内皮细胞。结论 此种脑微血管内皮细胞培养方法的建立为体外研究脑血管疾病提供了可靠的手段。
Objective To establish a simple and reliable method of culturing brain microvascular endothelial cells. Methods Primary cultured and subcultured rat cerebral microvascular endothelial cells were isolated from neonatal SD neonatal rat cortex by homogenization, filtration, digestion and differential adherent methods. The cultured cells were observed morphologically and cultured with Ⅷ factor-related antigen Immunocytochemistry was performed. Results The cultured cells showed monolayer adherent growth. Typical signs of “paving stones” appeared after about 7 to 9 days. Immunocytochemistry confirmed endothelial cells. Conclusion The establishment of this method of culturing brain microvascular endothelial cells provides a reliable means of studying cerebrovascular diseases in vitro.
