目的分析支架蛋白RACK 1在卵巢癌中表达的意义及其与基质金属蛋白酶-2(MMP-2)的相关性,探讨RACK1在卵巢癌发生中的可能作用机制。方法选取2009~2013年辽宁医学院附属第一医院手术切除的卵巢组织蜡块标本78例和正常卵巢组织10例,采用免疫组化PV法检测其RACK1和MMP-2的表达情况。结果 RACK 1在卵巢癌组织中的表达明显高于单纯性囊肿和卵巢良性组织(P<0.05);RACK1蛋白表达水平与卵巢癌转移和临床分期有关(P<0.05)。RACK1与MMP-2在卵巢癌中的表达存在相关性(r=0.357)。结论 RACK1在卵巢癌中的表达明显升高,与MMP-2在卵巢癌中的表达存在相关性,可能与肿瘤的发生发展、浸润、转移有密切关系。
Objective To analyze the significance of the expression of scaffold protein RACK 1 in ovarian cancer and its relationship with matrix metalloproteinase-2 (MMP-2) and to explore the possible mechanism of RACK1 in ovarian carcinogenesis. Methods Totally 78 cases of ovarian tissue paraffin blocks and 10 cases of normal ovarian tissue were collected from the First Affiliated Hospital of Liaoning Medical University from 2009 to 2013. The expression of RACK1 and MMP-2 were detected by immunohistochemistry. Results The expression of RACK 1 in ovarian cancer tissues was significantly higher than that in simple cysts and benign ovarian tissues (P <0.05). The expression of RACK1 protein was correlated with the metastasis and clinical stage (P <0.05). There was a correlation between RACK1 and MMP-2 expression in ovarian cancer (r = 0.357). Conclusions The expression of RACK1 in ovarian cancer is significantly increased, which is correlated with the expression of MMP-2 in ovarian cancer, which may be closely related to tumorigenesis, invasion and metastasis.
