目的探讨罗格列酮(Rosiglitazone,Ros)对同型半胱氨酸(homocysteine,Hcy)所致的人脐静脉内皮细胞(HUVECs)凋亡的影响。方法将原代培养的HUVECs分为对照组、Hcy刺激组和Ros干预组,MTT检测细胞存活率。免疫细胞化学方法检测Bax,Bcl-2,Caspase-3蛋白表达水平。采用Annexin V/PI染色加流式细胞仪检测细胞凋亡。比色法检测细胞培养上清液MDA和SOD含量。Western blot检测PPARγ蛋白表达水平。结果与对照组比较,Hcy组细胞存活率下降;Bax和Caspase-3表达上调,Bcl-2和PPARγ表达下调,MDA含量增加和SOD活性降低(P<0.05),而罗格列酮逆转上述变化(P<0.05)。结论罗格列酮可抑制Hcy所致的HUVECs凋亡。
Objective To investigate the effect of rosiglitazone (Ros) on the apoptosis of human umbilical vein endothelial cells (HUVECs) induced by homocysteine (Hcy). Methods Primary cultured HUVECs were divided into control group, Hcy stimulation group and Ros intervention group. The cell viability was detected by MTT assay. Immunocytochemistry was used to detect the protein expression of Bax, Bcl-2 and Caspase-3. Annexin V / PI staining and flow cytometry were used to detect apoptosis. Colorimetric assay of cell culture supernatant MDA and SOD content. Western blot detection of PPARγ protein expression levels. Results Compared with the control group, the cell viability of Hcy group was decreased; the expressions of Bax and Caspase-3, Bcl-2 and PPARγ were down-regulated, MDA content and SOD activity were decreased (P <0.05). Conclusion Rosiglitazone can inhibit Hcy-induced apoptosis of HUVECs.
