目的 检测糖皮质激素受体-α、p(GR-α、GR-β)在翼状胬肉组织中的表达,探讨其在翼状胬肉发病机制中的作用.方法 取39例翼状胬肉组织(其中初发30例,复发9例),应用荧光定量逆转录PCR和免疫组化技术检测GR-α、βmRNA和蛋白表达,并选择8例正常人的球结膜做为对照.结果 翼状胬肉及正常人的球结膜组织中均存在GR-αmRNA表达,但GR-β mRNA表达均为阴性.其中39例翼状胬肉中的GR-α mRNA为(10.40±1.42)× 10拷贝/μg,8例正常眼球结膜中GR-α mRNA为(2.23±0.32)×10拷贝/μg,差异有统计学意义(t=4.82,P0.05).结论 翼状胬内组织中GR-αmRNA及其蛋白表达明显增高,这可能是翼状胬肉发生、发展的重要原因之一,但其作用机制尚待于进一步研究.“,”Objective To investigate whether theglucocorticoid receptor would be expressed in pterygium and dis-cuss its significant. Methods GR- α and GR-β mRNA expression were detected by a real -time fluorescent quantitative RT- PCR. GR-α and GR-β protein expression were examined by Western- blotting analysis and immunohistochemicaltechnique from 39 patients with pterygium (30 primary pterygium, 9 recurrent pterygium). Normal conjunctivae(n =8) were as controls. Results The expression of GR -α nLRNA were (10. 40 ± 1. 42) × 10GR -α cDNA copies/μg total RNA in pterygium and (2.23 ±0. 32) × 10GR- α cDNA copies/μg total RNA in normal conjunctivae. The expression of GR- β mRNA was not detected in pterygium and in normal conjunctivae. There was no difference between the expression of GR -α mRNA in recurrent pterygium(10.50 ± 0.30)and the primary pterygium(10. 40 ± 0. 40)(P > 0. 05). There was significantly difference between the expression of GR - α mRNA in pterygium and the normal conjunctivae. Western - blotting analysis and immunohistochemical technique confirmed these findings for the protein transcripts of these genes. Conclusions These results demonstrate that the expression of GR-α mRNA and protein are higher in pterygium and probably are one of the important reasons. But its effection and meehanismin pterygium still need more investigate.
