目的探讨海生素对体外白血病K562细胞的抑制作用及机制。方法将0.1、1、10、100、1000μg/ml的海生素分别作用于K562细胞24、48 h,免疫细胞化学法检测凋亡相关蛋白bcl-2、bax的表达。结果海生素作用后K562细胞抑制率明显高于对照组,且呈现浓度及时间依赖性。K562细胞bcl-2蛋白表达下调,bax蛋白表达上调。结论海生素可抑制K562细胞的生长,其机制可能为通过下调bcl-2蛋白表达,增加bax蛋白的表达,诱导细胞凋亡。
Objective To investigate the inhibitory effect of seawater on K562 leukemia cells and its mechanism. Methods K562 cells were treated with 0.1, 1, 10, 100 and 1000μg / ml of seawater respectively for 24 and 48 h. The expressions of apoptosis related proteins bcl-2 and bax were detected by immunocytochemistry. Results The inhibitory rate of K562 cells was significantly higher than that of the control group after being treated with seawater. The concentration and time-dependent manner. The expression of bcl-2 protein was down-regulated and the protein expression of bax was up-regulated in K562 cells. Conclusions Sestatin can inhibit the growth of K562 cells, and its mechanism may be through down-regulating bcl-2 protein expression, increasing bax protein expression and inducing apoptosis.
