目的探讨脑室内出血后凝血酶对脑室周围组织是否存在毒性损伤作用。方法将凝血酶溶液注入鼠脑室内后,测定基底节与丘脑区域的脑血流量、血脑屏障通透性。另外,将鼠脑细胞置入含有凝血酶的培养液中培养,测定培养液中乳酸脱氢酶的浓度。结果凝血酶导致血脑屏障的破坏与脑细胞死亡,而对脑血流量没有明显影响。结论脑室内出血后,凝血酶对脑室周围组织存在毒性损伤,血脑屏障的破坏与细胞毒性作用可能是这一损伤的主要病理机制。
Objective To investigate the effect of thrombin on the periventricular tissue after intraventricular hemorrhage. Methods After the thrombin solution was injected into the mouse ventricle, the cerebral blood flow and the permeability of the blood-brain barrier in the basal ganglia and the thalamus were measured. In addition, rat brain cells were cultured in a culture broth containing thrombin, and the concentration of lactate dehydrogenase in the culture fluid was measured. Results Thrombin led to the destruction of the blood-brain barrier and brain cell death, but no significant effect on cerebral blood flow. Conclusion After intraventricular hemorrhage, thrombin has toxic injury to the periventricular tissues. The destruction of the blood-brain barrier and cytotoxicity may be the main pathological mechanism of this injury.
