目的:筛选适于酶解法生产降血压肽的水产蛋白原料。方法:在分析15种水产品与水产品下脚料基本成分的基础上,以酶解法(酸性蛋白酶和碱性蛋白酶)酶解15种原料,测定不同酶解液的水解度、ACE体外抑制率和ACEIC50。结果:碱性蛋白酶对原料的水解度较大;15种原料的酶解液均表现出不同的ACE抑制效果,抑制率在19%~99%之间;以ACEIC50为评价指标,ACE抑制效果好的原料有鲫鱼、鲈鱼、带鱼脊骨和鲳鱼,ACE-IC50分别为11.68、12.96、11.26、11.18mg/mL(酸性蛋白酶酶解)。结论:综合考虑降血压肽的工业化生产要素和ACE抑制活性,选取价格低廉、原料预处理简单的带鱼脊骨作为ACE抑制肽生产的水产蛋白原料。
Objective: To screen the aquatic protein raw materials suitable for the production of hypotensive peptide by enzymolysis. Methods: Based on the analysis of the basic components of 15 kinds of aquatic products and fishery products, 15 kinds of raw materials were digested by enzymatic hydrolysis (acid protease and alkaline protease), the degree of hydrolysis of different enzymatic hydrolyzate, the inhibition rate of ACE in vitro and ACEIC50. Results: The degree of hydrolysis of alkaline protease to raw materials was high. The ACE inhibitors of 15 kinds of raw materials all showed different inhibitory rates of ACE, with the inhibitory rates between 19% and 99%. ACE ACE50 was the better indicator of ACE inhibition The raw materials were crucian carp, perch, spine fish and anchovy, ACE-IC50 were 11.68,12.96,11.26,11.18mg / mL (acid protease digestion). Conclusion: Considering the industrial production factors and ACE inhibitory activity of hypotensive peptide, we chose the fishbone spine with low price and simple pretreatment as the ACE inhibitor for the production of aquatic protein raw materials.
