甜菊糖又称甜菊糖甙或甜菊素,其含量的测定在我国现行标准中采用容量法和高效液相色谱法。早期甜菊糖总甙中以甜菊甙为主成分,容量法测定含量也就建立在这一基础之上,但是近年来甜菊叶品种不断改良,其总甙主成分发生改变,瑞鲍迪甙A成为主成分,因此在实际检测中使用容量法所测结果与真实值便出现了偏差。试验通过高纯度甜菊甙和瑞鲍迪甙A单一成分按照不同比例混合,经容量法和高效液相色谱法分别测定其含量,结果表明随着瑞鲍迪甙A所占比例的增加,测定结果与真实值的偏差越大。经分析这可能与瑞鲍迪甙A分子质量较甜菊甙大有关,同等质量的瑞鲍迪甙A物质的量比甜菊甙要小,容量法滴定时消耗的碱液量就小,测定结果就会偏低。而高效液相色谱法就不会受此因素的影响,测定结果也更接近真实值。
