目的研究3′-大豆苷元磺酸钠促进人乳腺癌细胞MCF-7凋亡的作用。方法 3种人乳腺癌细胞系(MCF-7、MDA-MB-231和SK-BR-3)与不同浓度的3′-大豆苷元磺酸钠共孵育24 h,MTT法检测细胞存活率,流式细胞术和Hoechst 33258染色检测细胞凋亡。结果 3′-大豆苷元磺酸钠能够抑制3种人乳腺癌细胞系的生长,3′-大豆苷元磺酸钠对MCF-7、MDA-MB-231和SK-BR-3细胞的IC50分别为656、798、735μmol/L。Hoechst33258荧光染色与流式细胞术分析MCF-7细胞的结果一致,656、900μmol/L 3′-大豆苷元磺酸钠诱导MCF-7细胞凋亡率分别为(16.4%±3.2%)和(38.0%±4.1%)。结论 3′-大豆苷元磺酸钠抑制乳腺癌细胞MCF-7增殖并可诱导其凋亡。
Objective To study the effect of 3’-daidzein sulfonate on the apoptosis of human breast cancer cell line MCF-7. Methods Three human breast cancer cell lines MCF-7, MDA-MB-231 and SK-BR-3 were incubated with different concentrations of 3’-daidzein sulfonate for 24 h. Cell viability was measured by MTT assay. Flow cytometry and Hoechst 33258 staining were used to detect apoptosis. Results Sodium 3’-daidzein sulfonate inhibited the growth of 3 human breast cancer cell lines, and the IC50 of 3’-daidzein sulfonate on MCF-7, MDA-MB-231 and SK-BR-3 cells Respectively 656,798,735μmol / L. Hoechst33258 fluorescence staining and flow cytometry analysis of MCF-7 cells results, 656,900μmol / L 3’-daidzein sulfonate induced apoptosis rate of MCF-7 cells were (16.4% ± 3.2%) and 38.0% ± 4.1%). Conclusion 3’-daidzein sulfonate inhibits the proliferation and induces the apoptosis of breast cancer cell MCF-7.
