目的:探讨无患子皂苷对人肺癌A549细胞的影响及其可能的分子机制。方法:采用MTT法检测无患子皂苷溶液对A549细胞增殖抑制作用;Hoechst 33258荧光染色和流式细胞术观察和检测细胞凋亡情况;实时荧光定量PCR法检测凋亡基因bcl-2 m RNA的表达。结果:无患子皂苷能显著抑制A549细胞的增殖,且呈浓度依赖性;Hoechst 33258荧光染色和流式细胞术结果显示无患子皂苷可以诱导A549细胞凋亡,实时荧光定量PCR法表明不同浓度的无患子皂苷均能明显降低bcl-2的表达。结论:无患子皂苷可通过诱导细胞凋亡而抑制A549细胞增殖,其作用可能与下调细胞凋亡相关基因bcl-2的表达有关。
Objective: To investigate the effect of Saponin on human lung cancer A549 cells and its possible molecular mechanism. Methods: The inhibitory effect of Saponin on proliferation of A549 cells was detected by MTT assay. The apoptosis of A549 cells was detected by Hoechst 33258 staining and flow cytometry. The apoptosis rate of bcl-2 mRNA was detected by real-time fluorescence quantitative PCR expression. Results: Saponins of Saponins could significantly inhibit the proliferation of A549 cells in a concentration-dependent manner. The results of Hoechst 33258 staining and flow cytometry showed that Sapindus saponin could induce the apoptosis of A549 cells. Real-time fluorescence quantitative PCR showed that different concentrations Sapindus saponin can significantly reduce the expression of bcl-2. Conclusion: Sapindus saponins can inhibit the proliferation of A549 cells by inducing apoptosis, which may be related to the down-regulation of the expression of bcl-2.
