通过单因素和正交试验对秦岭细粘束孢分泌纤维素酶的组分、条件等进行研究。结果表明:秦岭细粘束孢可以分泌内切葡萄糖苷酶(CX)、外切葡萄糖苷酶(C1)和β-葡萄糖苷酶(BG),具有完整的纤维素酶系;其中CX是秦岭细粘束孢分泌的主要纤维素酶组分,优化后CX酶的最大酶活为35.215U,分别是C1和BG最大酶活的13.662倍和8.780倍。正交试验表明:秦岭细粘束孢分泌CX,C1和BG的最适条件各不相同,产CX的最佳条件为29℃、6天、起始pH4.0、摇床转速150r.min-1。温度、转速、起始pH和时间4个因素对秦岭细粘束孢产3种酶的影响各不相同,影响最大的是温度,其次是转速和时间,而起始pH对该菌产酶的影响最小。
Through single factor and orthogonal test, the components and conditions of cellulase secreted by Ceratophyllum demersum in Qinling Mountains were studied. The results showed that C. ceratosa could secrete endo-glucosidase (CX), exo-glucosidase (C1) and beta-glucosidase (BG) with complete cellulase system; among them, CX was fine The main cellulase component secreted by Myzus persicae was 35.215U, which was 13.662 and 8.780 times of the maximum activity of C1 and BG, respectively. The results of orthogonal test showed that the optimal conditions for the secretion of CX, C1 and BG were all different. The optimal conditions for producing CX were 29 ℃, 6 days, initial pH4.0, shaking speed 150r.min- 1. The effects of temperature, rotation speed, initial pH and time on the three kinds of enzymes of Myzus persicae were different, the most significant ones were temperature, followed by speed and time, Minimal impact.
