桃红四物汤对高同型半胱氨酸血症大鼠主动脉死亡受体途径凋亡的干预作用

目的:观察桃红四物汤对高同型半胱氨酸大鼠主动脉死亡受体途径凋亡的影响。方法:40只雄性SD大鼠随机分为正常组、模型组、桃红组、叶酸组,每组10只,正常组给予普通饲料,模型组、桃红组、叶酸组给予高蛋氨酸饮食(普通饲料添加2%蛋氨酸)喂养,桃红组采用桃红四物汤水煎液,按4.59 g生药/(kg.d)灌胃;叶酸组采用叶酸混悬液,按1.35 mg/(kg.d)灌胃;模型组及正常组给双蒸水2 mL/(只.d)灌胃。12周后,Elisa法检测血清hcy、TNF-α、MCP-1浓度,免疫组化检测主动脉Cleaved Caspase3表达,RT-PCR法检测主动脉TNF-α、Fas mRNA表达。免疫印迹法检测主动脉TNF-α的表达。结果:12周后,模型组大鼠血Hcy显著升高,血清TNF-α浓度升高,血清MCP-1浓度有升高趋势;主动脉内皮细胞连续性受损,受损部位下的血管平滑肌细胞Cleaved Caspase3呈片状黄褐色阳性染色。主动脉全层行RT-PCR示TNF-α、Fas mRNA表达上调,western blot示TNF-α表达上调。相对于模型组,桃红组和叶酸组大鼠血清TNF-α浓度降低,主动脉未发现片状Cleaved Caspase3阳性染色,TNF-αmRNA表达明显下调,Fas mRNA表达有下调趋势,TNF-α蛋白表达下调。但叶酸组伴有血同型半胱氨酸浓度的下降,而桃红组没有降低血同型半胱氨酸浓度的作用。结论:高同型半胱氨酸血症诱导SD大鼠主动脉死亡受体途径凋亡,桃红四物汤可降低血清TNF-α,并在基因和蛋白水平下调主动脉TNF-α表达。 Objective: To observe the effect of Taohongsiwutang on the apoptosis of aortic death receptor in hyperhomocysteine ​​rats. Results: After 12 weeks, the level of blood Hcy in the model group was significantly increased, the level of serum TNF-α was increased and the concentration of serum MCP-1 was increased. The aortic endothelial cells were damaged continuously, the vascular smooth muscle Cells Cleaved Caspase3 was flaky brown positive staining. Full-thickness aorta line RT-PCR showed TNF-α, Fas mRNA expression was up-regulated, western blot showed TNF-α upregulation. Compared with the model group, serum TNF-αlevel in the rosuvastatin group and folate-treated rats decreased, but in the aorta, no patchy Cleaved Caspase3 positive staining was found, the expression of TNF-αmRNA was down-regulated, the expression of Fas mRNA was down-regulated and the expression of TNF- . But folic acid group with blood homocysteine ​​concentration decreased, while the pink group did not reduce the concentration of blood homocysteine. CONCLUSION: Hyperhomocysteinemia induces apoptosis of aortic death receptors in SD rats. Taohong Siwu decoction can decrease serum TNF-α and down-regulate the expression of TNF-α in the aorta at gene and protein level.
