目的:观察清上蠲痛合剂对偏头痛大鼠中脑P物质mRNA表达的影响。方法:将成年健康SD大鼠84只,雌雄各半,随机分为正常组,模型组,清上蠲痛合剂预防组,清上蠲痛合剂高、中、低剂量治疗组,舒马普坦组,每组12只。采用硝酸甘油(GTN)皮下注射法建立偏头痛大鼠模型,给药结束后,取脑组织,采用RT-PCR检测SP mRNA表达量。结果:与正常组相比,模型组大鼠中脑P物质mRNA表达显著降低(P≤0.01)。舒马普坦阳性组、清上蠲痛合剂预防组、高、中剂量治疗组大鼠中脑P物质mRNA表达上升(P<0.05),与模型组比较有统计学意义(P<0.05)。清上蠲痛合剂预防组、治疗组高、治疗组中、舒马普坦阳性组之间无统计学意义(P>0.05)。结论:清上蠲痛合剂能明显预防和改善大鼠偏头痛发作,可能与上调偏头痛大鼠中脑P物质mRNA表达有关。
Objective: To observe the effect of Qing Shang Tiao Tong Mixture on substance P mRNA expression in midbrain of migraine rats. Methods: 84 adult healthy SD rats were randomly divided into normal group, model group, Qing Shang Tu Kou Mixture Prevention group, Qing Shang Tu Kao Mixture high, medium and low dose treatment group, sumatriptan Group, 12 in each group. The migraine rat model was established by subcutaneous injection of nitroglycerin (GTN). After administration, brain tissue was taken and the expression of SP mRNA was detected by RT-PCR. Results: Compared with the normal group, the expression of substance P mRNA in the midbrain decreased significantly in the model group (P≤0.01). Sumatriptan positive group, Qing Shangchong Tong mixture prevention group, high and medium dose treatment group rats brain substance P mRNA expression increased (P <0.05), compared with the model group was statistically significant (P <0.05). There were no significant differences between the two groups (P> 0.05). Conclusion: Qing Shang Tiao Tong Mixture can obviously prevent and improve migraine attacks in rats, which may be related to up-regulation of substance P mRNA in the midbrain of migraine rats.
