目的 研究COX - 2和iNOS在大肠腺癌中的表达和相互作用 ,其及它们与大肠腺癌生物学行为的关系。方法 采用SP免疫组化方法检测了 76例手术切除的大肠腺癌标本和 15例正常肠粘膜内COX - 2和iNOS蛋白的表达。结果 COX - 2和iNOS在大肠腺癌中阳性率 (76 .3%、85 .5 % )显著高于正常大肠粘膜(13.3%、2 0 % ,P <0 .0 0 5 ) ;不同组织类型大肠腺癌之间COX - 2阳性积分有显著差异 (P <0 .0 0 5 ) ,乳头状腺癌、管状腺癌Ⅰ级和管状腺癌Ⅱ级显著高于管状腺癌Ⅲ级 (P <0 .0 0 5 )和粘液腺癌 (P <0 .0 0 5 ) ;有淋巴结转移和无淋巴结转移者COX - 2的表达无显著差异 (P >0 .0 5 ) ;不同组织类型大肠腺癌之间iNOS阳性积分无显著差异 (P >0 .0 5 ) ;有淋巴结转移组iNOS的表达显著高于无淋巴结转移组 (P <0 .0 1) ;大肠腺癌组织中COX- 2和NOS的表达无相关性 (χ2 =0 .15 35 ;P >0 .0 5 )。结论 COX - 2和iNOS过度表达参与了大肠腺癌的发生展过程 ,并与其生物学行为有关 ;在大肠腺癌中COX - 2和iNOS的表达无相关性 ,提示它们在大肠腺癌的发生发展中所起的作用不同 ,是两个相互独立的作用因子。
Objective To study the expression and interaction of COX - 2 and iNOS in colorectal adenocarcinoma and their relationship with the biological behavior of colorectal adenocarcinoma. Methods The expression of COX - 2 and iNOS protein in 76 cases of resected colorectal adenocarcinoma and 15 cases of normal intestinal mucosa were detected by SP immunohistochemistry. Results The positive rate of COX - 2 and iNOS in colorectal adenocarcinoma was significantly higher than that in normal colorectal mucosa (76.3%, 85.5%) (13.3%, 20%, P <0.05) There were significant differences in COX - 2 positive scores between colorectal adenocarcinoma (P <0.05), papillary adenocarcinoma, tubular adenocarcinoma Ⅰ and tubular adenocarcinoma Ⅱ, which were significantly higher than those of tubular adenocarcinoma Ⅲ (P < 0.05). There was no significant difference in the expression of COX - 2 between lymph node metastasis and non - lymph node metastasis (P> 0.05). The expression of COX - 2 in different tissue types There was no significant difference in iNOS positive score between the two groups (P> 0.05). The expression of iNOS in lymph node metastasis group was significantly higher than that in non-lymph node metastasis group (P <0.01) NOS expression was not related (χ2 = 0.1535; P> 0.05). Conclusions Overexpression of COX - 2 and iNOS is involved in the process of colorectal adenocarcinoma development and is related to their biological behavior. There is no correlation between the expression of COX - 2 and iNOS in colorectal adenocarcinoma, suggesting that they may play an important role in the development of colorectal adenocarcinoma Play a different role in the role of the two independent factors.
