目的用全细胞膜片钳技术和 Western 印迹法,探讨急性冠状动脉综合征(ACS)患者外周血淋巴细胞电压依赖性钾通道(Kv)电流及 Kv1.3蛋白表达的变化。方法收集12例 ACS 患者和10例健康志愿者外周血淋巴细胞,采用膜片钳全细胞电流记录方法,记录淋巴细胞膜 Kv 的电流密度。用 Western 印迹法检测外周血淋巴细胞 Kv1.3蛋白的表达。结果 ACS 患者淋巴细胞 Kv 的电流密度为(269±94)pA/pF,明显高于正常人[(191±64)pA/pF,P<0.01]。ACS 组淋巴细胞膜电容为(2.3±0.7)pF,对照组为(2.2±0.5)pF,两组间差异无统计学意义。Western 印迹结果显示 ACS 患者淋巴细胞 Kv1.3通道表达高于正常人(P<0.01)。结论 ACS 患者淋巴细胞 Kv 表达明显增多,说明 Kv 在调节 ACS 患者淋巴细胞的激活中起着关键性作用。
Objective To investigate the changes of Kv current and Kv1.3 protein expression in peripheral blood lymphocytes in patients with acute coronary syndrome (ACS) by whole-cell patch clamp technique and Western blotting. Methods Peripheral blood lymphocytes were collected from 12 patients with ACS and 10 healthy volunteers. Whole-cell patch clamp current recording was used to record the current density of lymphocytes Kv. Western blotting was used to detect the expression of Kv1.3 protein in peripheral blood lymphocytes. Results The current density of Kv in lymphocytes was (269 ± 94) pA / pF in ACS patients, which was significantly higher than that in normal controls [(191 ± 64) pA / pF, P <0.01]. The membrane capacitance of lymphocytes was (2.3 ± 0.7) pF in ACS group and (2.2 ± 0.5) pF in control group, with no significant difference between the two groups. Western blot results showed that the expression of Kv1.3 channel of lymphocytes in ACS patients was higher than that in normal people (P <0.01). Conclusions The expression of Kv in lymphocytes of patients with ACS is significantly increased, indicating that Kv plays a key role in the activation of lymphocytes in patients with ACS.
