探讨了不同胚龄鸡胚旋转不同时间后,脑细胞内游离Ca2+水平的变化。利用回转器模拟微重力生物效应及fura-2比例荧光法,测定了孵化第6至18d鸡胚脑细胞游离Ca2+的浓度。结果显示:孵化第8至17d鸡胚经旋转不同时间后,其脑细胞内游离Ca2+浓度均有所下降,其中第10d鸡胚旋转4或7h及第13d鸡胚旋转24h后,脑细胞内Ca2+浓度比对照组显著降低(P<0.01)。第10d和13d鸡胚分别旋转4h和24h后再静置恢复相同时间,细胞内Ca2+浓度均有所增加,但仍低于对照组水平。结果表明:回转器旋转引起的鸡胚脑细胞内游离Ca2+水平降低是可逆的,而且在鸡胚脑后部的红色光灸,对鸡胚脑细胞内Ca2+水平降低的恢复具有一定促进作用
The changes of intracellular free Ca2 + levels in different time of embryo chicken embryos were investigated. The cyclotron was used to simulate the effects of microgravity and the fura-2 ratio fluorescence method was used to determine the concentration of free Ca2 + in chicken embryo brain cells from the 6th to the 18th day after hatching. The results showed that after 8 to 17 days of incubation, the free Ca2 + concentration in brain cells decreased at different time points. After 4 or 7 hours of embryo rotation and 24 hours of embryo rotation at 13 days, the intracellular Ca2 + The concentration was significantly lower than the control group (P <0.01). At the 10th and 13th day, the chick embryos were respectively rotated for 4h and 24h, and then resumed for the same time. The intracellular Ca2 + concentration was increased but still lower than that of the control group. The results showed that the decrease of intracellular free Ca2 + level in chicken embryo brain cells induced by rotator rotation was reversible, and the red light moxibustion in the posterior portion of chick embryo had a certain promotion effect on the reduction of intracellular Ca2 + level in chicken embryos
