目的利用基因芯片技术检测增生期与消退期血管瘤中血管生成相关基因表达的差异。探讨血管瘤增生与自然消退的机制。方法增生期和消退期血管瘤瘤体组织各4例。抽提瘤体mRNA,逆转录为cDNA,制备成cDNA探针。将cDNA探针与SuperArray公司产人血管生成基因芯片(可检测基因111条)杂交,通过计算机扫描、数据处理后筛选出表达有差异的基因。结果表达有差异的基因共59条,其中增生期血管瘤有30条基因表达上调,消退期血管瘤有29条基因表达上调。差异表达基因包括一些特定启动子和抑制子、生长因子及其受体、细胞因子和化学因子以及粘附分子、细胞外基质蛋白及转录因子等基因。结论血管瘤的增生、消退和多个血管生成相关基因的差异表达有关。
Objective To detect the difference of angiogenesis-related gene expression in proliferative and regression hemangiomas by using gene chip technique. To investigate the mechanism of hemangiomas proliferation and spontaneous regression. Methods Hemangiomas in proliferative phase and regressive phase each had 4 cases. Tumor mRNA was extracted, reverse transcribed into cDNA, prepared as a cDNA probe. The cDNA probes were hybridized with SuperArray’s human angiogenesis gene chip (111 detectable genes) and screened by computer. After data processing, the differentially expressed genes were screened out. Results There were 59 differentially expressed genes, of which 30 genes were up-regulated in proliferating hemangiomas and 29 genes were up-regulated in cancerous hemangiomas. Differentially expressed genes include a number of specific promoters and suppressors, growth factors and their receptors, cytokines and chemokines and adhesion molecules, extracellular matrix proteins and transcription factors and other genes. Conclusions The proliferation, regression of hemangiomas and the differential expression of multiple angiogenesis related genes are related.
