目的:阐明胃癌生长对氨基酸代谢的影响以及对某些氨基酸的特殊需求。 方法:采用RP-HPLC技术检测12例胃癌病人肿瘤组织20 种游离氨基酸含量,且与肿瘤体积进行相关性和多元逐步回归分析。 结果:胃癌组织游离谷氨酰胺、精氨酸、蛋氨酸含量较正常胃粘膜明显增高(P< 0.05),而丙氨酸、色氨酸含量较正常胃粘膜明显降低(P< 0.05)。同时胃癌组织游离谷氨酰胺、精氨酸、蛋氨酸含量与肿瘤体积呈正相关,而丙氨酸与肿瘤体积呈负相关。 结论:随着胃癌生长,肿瘤细胞特别需求谷氨酰胺、蛋氨酸、精氨酸作为蛋白质、核酸合成的原料,因而导致氨基酸代谢的改变
Objective: To elucidate the effects of gastric cancer growth on amino acid metabolism and the specific needs of certain amino acids. Methods: RP-HPLC was used to detect the contents of 20 free amino acids in tumor tissues of 12 patients with gastric cancer, and the correlation with tumor volume and multivariate stepwise regression analysis. Results: The contents of free glutamine, arginine, and methionine in gastric cancer tissue were significantly higher than those in normal gastric mucosa (P<0.05). The content of alanine and tryptophan in gastric cancer tissue was significantly lower than that in normal gastric mucosa (P<0.05). At the same time, the content of free glutamine, arginine, and methionine in gastric cancer tissues was positively correlated with tumor volume, while alanine was negatively correlated with tumor volume. Conclusion: With the growth of gastric cancer, tumor cells especially require glutamine, methionine, and arginine as raw materials for protein and nucleic acid synthesis, resulting in changes in amino acid metabolism.
