目的:探索雌激素受体阴性(ER-)Ⅱ期乳腺浸润性导管癌的蛋白标志物,阐述获得蛋白在肿瘤浸润、转移方面的意义。方法:利用二维凝胶电泳(2-DE)、基质辅助激光解吸电离飞行时间质谱技术(MALDI-TOF-MS)及在线数据库检索,对5例Ⅱ期乳腺浸润性导管癌ER-癌组织及癌旁组织的差异表达蛋白质进行鉴定分析。结果:5例癌旁正常乳腺组织中检测出的蛋白点数从779到1328不等,实体瘤组织中检测出的蛋白点数从988到1453不等,两者间的匹配点从310到774不等。5例患者乳腺癌组织和癌旁正常乳腺组织的全蛋白差异表达凝胶图谱上确定3个差异蛋白质点S1、S2和S3,鉴定出3个候选蛋白:免疫球蛋白λ链可变区(S1)、胶原蛋白Ⅵ-3链前体(S2)及S100A11钙结合蛋白(S3)。其中S2和S3在5例癌组织中表达均上调(丰度差异2倍以上)。结论:优化的实体瘤组织二维电泳实验条件能够从实体瘤组织中更有效地提取蛋白质;胶原蛋白Ⅵ-3链前体及S100A11钙结合蛋白可能与ER-Ⅱ期乳腺浸润性导管癌的浸润及转移有关。
Objective: To explore the protein markers of estrogen receptor-negative (ER-) stage Ⅱ breast invasive ductal carcinoma and to elucidate the significance of the obtained proteins in tumor invasion and metastasis. Methods: Five cases of invasive ductal carcinoma of the breast (ERCC) from 5 cases of stage Ⅱ breast cancer were treated with two-dimensional gel electrophoresis (2-DE), matrix-assisted laser desorption / ionization time of flight mass spectrometry (MALDI-TOF-MS) and online database search Paracancerous tissue differentially expressed proteins were identified and analyzed. Results: The number of protein spots detected in 5 cases of adjacent normal breast tissue ranged from 779 to 1328, and the number of detected protein spots in solid tumor tissues ranged from 988 to 1453. The matching points ranged from 310 to 774 . Three different protein spots S1, S2 and S3 were identified on the differentially expressed gel map of breast cancer tissues and adjacent normal breast tissues in 5 patients. Three candidate proteins were identified: immunoglobulin λ chain variable region (S1 ), Collagen VI-3 chain precursor (S2) and S100A11 calcium binding protein (S3). Among them, S2 and S3 were all upregulated in 5 cases of cancerous tissues (more than 2 folds difference in abundance). CONCLUSION: The optimized two-dimensional electrophoresis of solid tumor tissue can extract protein from solid tumor tissue more effectively. Collagen VI-3 chain precursor and S100A11 calcium-binding protein may be involved in the infiltration of ER-Ⅱ breast invasive ductal carcinoma And transfer related.
