目的:观察高糖条件下大鼠肾小球系膜细胞(GMC)分泌细胞外基质(ECM)成分中纤粘连蛋白(FN)、层粘连蛋白(LAM)、C-Ⅲ(Ⅲ型胶原)和C-Ⅳ(Ⅳ型胶原)的变化。方法:采用体外大鼠肾小球系膜细胞培养,用ELISA方法进行有关测定。结果:高糖对体外培养的系膜细胞分泌ECM的影响具有选择性,仅增加功能相关蛋白的合成,而对其他类型的间质胶原没有影响。结论:体外高糖可增加细胞外基质蛋白的合成。由此可以推断,在体内肾小球系膜细胞对高血糖的反应是增加ECM的合成,加速ECM的积聚,从而导致糖尿病肾小球硬化。
OBJECTIVE: To observe the changes of fibronectin (FN), laminin (LAM), C-Ⅲ (collagen Ⅲ) and extracellular matrix (EC) in rat glomerular mesangial cells C-Ⅳ (type Ⅳ collagen) changes. Methods: Rat mesangial cells were cultured in vitro and assayed by ELISA. Results: The effect of high glucose on the secretion of ECM in cultured mesangial cells was selective, only increased the synthesis of functionally related proteins, but had no effect on other types of interstitial collagen. Conclusion: In vitro high glucose can increase the extracellular matrix protein synthesis. From this we can conclude that the response of glomerular mesangial cells to hyperglycemia in vivo is to increase the synthesis of ECM and accelerate the accumulation of ECM, leading to diabetic glomerulosclerosis.
