目的:探究胆石六号汤剂对胆色素结石豚鼠胆囊收缩素A受体(CCKAR)、5-羟色胺3a受体(5-HT3aR)表达的影响,从而为胆石六号汤剂的临床使用提供实验依据。方法:将40只雌性豚鼠随机分为空白组(n=10)、模型组(n=15)和胆石六号汤剂组(n=15),除空白组外,其余2组采用饮食诱发法建立胆色素结石模型。胆石六号汤剂组给予胆石六号汤剂7.7 g·kg-1灌胃治疗。30 d后观察各组的成石率、胆囊CCKAR、5-HT3aR基因和蛋白的表达情况。结果:模型组成石率为91.67%,显著高于空白组10.00%和胆石六号汤剂组26.67%。RT-PCR检测模型组中CCKAR、5-HT3aR基因的表达明显低于空白组和胆石六号汤剂组,差异有统计学意义(P<0.05)。免疫组化结果显示模型组5-HT3aR蛋白相对于空白组显著下降,而胆石六号汤剂组阳性表达相对于模型组显著上升,差异有统计学意义(P<0.05)。模型组CCKAR蛋白的表达相对于空白组有所下降,胆石六号汤剂组阳性表达相对于模型组有上升趋势,但是差异没有统计学意义(P>0.05)。结论:胆石六号汤剂显著影响了胆色素结石豚鼠胆组织CCKAR、5-HT3aR基因及蛋白的表达,其治疗胆色素结石的作用一定程度上是通过增加它们的表达来实现的。
Objective: To investigate the effect of Ganshuang No.6 decoction on the expression of cholecystokinin receptor (CCKAR) and 5-HT3aR in guinea pigs with bile pigment stones, so as to provide experimental data for the clinical use of gallstone 6 decoction in accordance with. Methods: Forty female guinea pigs were randomly divided into blank group (n = 10), model group (n = 15) and gallstone 6 decoction group (n = 15) To establish a model of bile pigment stones. Ganshuang No. 6 decoction group was given gutase No. 6 decoction 7.7 g · kg-1 intragastrically. After 30 days, the rate of stone formation, expression of CCKAR and 5-HT3aR gene and protein in gallbladder were observed. Results: The stone composition rate of the model was 91.67%, which was significantly higher than that of the control group (10.00%) and that of the Dan Shi Liu Decoction group (26.67%). The expression of CCKAR and 5-HT3aR genes in RT-PCR assay group was significantly lower than that in blank group and GST group. The difference was statistically significant (P <0.05). The results of immunohistochemistry showed that the expression of 5-HT3aR protein in the model group was significantly decreased compared with the blank group, while the expression level in the gallstone Liuhuangji Decoction group was significantly increased compared with the model group (P <0.05). The expression of CCKAR protein in the model group decreased compared with that in the blank group. The positive expression of CCKAR in the Jintan Liu decoction group increased compared with the model group, but the difference was not statistically significant (P> 0.05). CONCLUSION: Ganshuang No.6 Decoction significantly affects the expression of CCKAR and 5-HT3aR gene and protein in the bile tissue of guinea pigs with bile pigment stones. The treatment of bile pigment stones can be achieved by increasing their expression to a certain extent.
