目的观察氯沙坦对自发性高血压大鼠肾脏结缔组织生长因子表达的调控。方法 14周龄雄性自发性高血压大鼠(SHR)随机分为高血压组和氯沙坦组,各8只,14周龄雄性Wistar-kyoto大鼠8只为正常对照组(WKY组),氯沙坦组给予氯沙坦30mg/(kg.d),16周后,观察各组大鼠体质量、收缩压,免疫组化和RT-PCR检测肾脏结缔组织生长因子(CTGF)、Ⅲ型胶原的表达。结果①高血压组肾脏CTGF、Ⅲ型胶原表达明显高于WKY组(P<0.05);氯沙坦组肾脏CTGF、Ⅲ型胶原表达较高血压组明显降低(P<0.05);②CTGFmRNA和蛋白表达量与Ⅲ型胶原mRNA和蛋白表达量显正相关(r1=0.522,r2=0.603,均P<0.01)。结论氯沙坦可能通过抑制SHR肾脏结缔组织生长因子表达,对早期高血压肾间质损害有保护作用。
Objective To observe the effects of losartan on the expression of connective tissue growth factor in spontaneously hypertensive rats. Methods 14-week-old male spontaneously hypertensive rats (SHR) were randomly divided into hypertensive group and losartan group, 8 rats in each group. Eight male Wistar-kyoto rats, 14 weeks old, were randomly divided into normal control group (WKY group) Losartan 30mg / (kg.d) was given in losartan group, and after 16 weeks, the body weight, systolic pressure, the expression of connective tissue growth factor (CTGF), type Ⅲ Collagen expression. Results ① The expression of CTGF and type Ⅲ collagen in renal tissue of hypertension group was significantly higher than that of WKY group (P <0.05). The expression of CTGF and Ⅲ collagen in losartan group was significantly lower than that of hypertension group (P <0.05); ② The expression of CTGF mRNA and protein The amount of type Ⅲ collagen mRNA and protein expression was positively correlated (r1 = 0.522, r2 = 0.603, both P <0.01). Conclusion Losartan may protect the early hypertensive renal interstitium by inhibiting the expression of connective tissue growth factor in SHR kidney.
