目的探讨miRNA-183下调Ezrin蛋白表达对人成骨肉瘤细胞系MG-63细胞侵袭转移能力的影响。方法以qRT-PCR和Western blot技术检测人成骨肉瘤细胞系MG-63细胞在转染miRNA-183模拟物前后miRNA-183和Ezrin蛋白的相对表达量。同时以划痕实验和transwell实验分析转染miRNA-183模拟物对MG-63细胞迁移、侵袭能力的影响。结果与正常人胚成骨细胞hFOB细胞相比,人成骨肉瘤细胞MG-63存在miRNA-183的低表达和Ezrin蛋白的表达上调(P<0.05)。在稳定转染miRNA-183模拟物后,MG-63细胞miRNA-183丰度显著上调,并明显抑制Ezrin蛋白的表达(P<0.05)。而转染后的MG-63细胞与转染前相比,其迁移和侵袭能力也显著降低(P<0.05)。结论 MiRNA-183可以通过下调靶蛋白Ezrin表达抑制人成骨肉瘤细胞系MG-63细胞的侵袭转移能力。
Objective To investigate the effect of miRNA-183 down-regulating Ezrin protein expression on invasion and metastasis of human osteosarcoma cell line MG-63. Methods The relative expression levels of miRNA-183 and Ezrin in human osteosarcoma cell line MG-63 before and after transfection of miRNA-183 mimics were detected by qRT-PCR and Western blot. Scratch assay and transwell assay were used to analyze the effect of miRNA-183 mimics on the migration and invasion of MG-63 cells. Results Compared with normal human embryonic osteoblast hFOB cells, the expression of miRNA-183 and Ezrin protein were up-regulated in human osteosarcoma MG-63 cells (P <0.05). After stable transfection of miRNA-183 mimics, the miRNA-183 abundance of MG-63 cells was significantly up-regulated and the expression of Ezrin protein was significantly inhibited (P <0.05). However, the ability of migration and invasion of transfected MG-63 cells was significantly lower than that before transfection (P <0.05). Conclusion MiRNA-183 can inhibit the invasion and metastasis of human osteosarcoma cell line MG-63 by down-regulating the expression of Ezrin.
