六味地黄丸对庆大霉素肾中毒大鼠肾核因子κB的影响

目的:观察六味地黄丸对大鼠庆大霉素肾毒性的疗效并初步探索其作用机制。方法:将大鼠随机分为3组:正常对照组(C)、模型组(GM)、六味地黄丸组(LDW)。采用生化检测和蛋白免疫印迹方法观察各组动物肾功能、氧化产物及酶类的变化及用药对信号通路分子p38、核因子κB(NF-κB)蛋白表达的影响。结果:与正常对照组比较,GM组大鼠体质量显著下降,肾脏指数、肾功能、肾皮质氧化产物MDA含量与NOS酶活性显著增高,SOD酶活性显著降低,NF-κB蛋白表达显著增多。与GM组比较,治疗第1周末LDW组大鼠肾功能、肾皮质NOS活性显著降低,SOD活性显著增高,NF-κB蛋白表达显著降低。结论:六味地黄丸能通过调节NF-κB的表达改善庆大霉素所致肾脏氧化损伤,对庆大霉素肾中毒具有一定疗效。 Objective: To observe the effect of Liuweidihuangwan on gentamicin nephrotoxicity in rats and to explore its mechanism. Methods: The rats were randomly divided into 3 groups: normal control group (C), model group (GM) and Liuweidihuangwan group (LDW). The changes of renal function, oxidized products and enzymes in each group were observed by biochemical detection and Western blotting, and the effect of drugs on the expression of signal transduction molecules p38 and nuclear factor kappa B (NF-κB) was examined. Results: Compared with the normal control group, the body weight of rats in the GM group decreased significantly. The renal index, renal function, MDA content in the renal cortex and NOS activity were significantly increased. The activity of SOD was significantly decreased and the expression of NF-κB was significantly increased. Compared with the GM group, renal function and renal cortex NOS activity in the LDW group were significantly decreased, SOD activity was significantly increased and the expression of NF-κB protein was significantly decreased in the first weekend. Conclusion: Liuweidihuangwan can improve oxidative damage of gentamicin-induced kidney by regulating the expression of NF-κB, and has a certain effect on gentamicin-induced nephrotoxicity.
