目的进行不同品系蚊转铁蛋白基因表达量的分析。方法据二维电泳获得肽段序列设计引物扩增基因片段,荧光定量PCR检测在淡色库蚊抗性品系品系、敏感品系、现场采集品系蚊体内的表达水平差异。结果成功扩增淡色库蚊转铁蛋白基因片段,荧光定量PCR表明转铁蛋白基因在抗性品系蚊体内的表达量是敏感品系的13.52倍,现场品系是敏感品系的7.48倍。结论淡色库蚊转铁蛋白基因可做为新的抗性检测及治理基因靶标。
Objective To analyze the gene expression of mosquito transferrin in different strains. Methods Two-dimensional electrophoresis was used to amplify the gene fragment by primer design. Fluorescent quantitative PCR was used to detect the expression level of the genes in the resistant, susceptible and field mosquitoes of Culex pipiens pallens. Results The fragment of transferrin gene of Culex pipiens pallens was successfully amplified. Fluorescence quantitative PCR showed that the expression level of transferrin gene in resistant mosquito was 13.52 times of that in susceptible strain, and 7.48 times in the susceptible strain. Conclusion Culex pterodactylum transferrin gene can be used as a new gene detection and control gene target.
