目的研究CD147 siRNA转染对恶性黑素瘤细胞株A375中CD147表达及细胞增殖的影响。方法将前期构建的重组质粒pSUPER/CD147 siRNA稳定转染至恶性黑素瘤细胞株A375中,采用半定量RT-PCR法检测转染细胞中CD147 mRNA的表达。采用MTT法检测转染pSUPER/CD147 siRNA对肿瘤细胞增殖的影响。结果转染了pSUPER/CD147 siRNA的恶性黑素瘤细胞株A375中CD147 mRNA 表达水平显著下调,其在转染后24 h、48 h和72 h的增殖水平均显著下降。结论 siRNA转染能有效抑制恶性黑素瘤细胞株A375中CD147的表达,且能抑制肿瘤细胞的增殖能力。
Objective To investigate the effect of CD147 siRNA transfection on CD147 expression and cell proliferation in malignant melanoma cell line A375. Methods The recombinant plasmid pSUPER / CD147 siRNA was stably transfected into A375 cell line. The expression of CD147 mRNA in transfected cells was detected by semi-quantitative RT-PCR. The effect of pSUPER / CD147 siRNA transfection on tumor cell proliferation was detected by MTT assay. Results The expression of CD147 mRNA was significantly down-regulated in the transfected pSUPER / CD147 siRNA-transfected A375 cells. The proliferation of CD147 mRNA was significantly decreased at 24 h, 48 h and 72 h after transfection. Conclusion siRNA transfection can effectively inhibit the expression of CD147 in malignant melanoma cell line A375 and inhibit the proliferation of tumor cells.
