以植物组织培养技术建立的9种晋产北柴胡(Bupleurum chinense DC.)试管植株为材料,用CTAB法提取北柴胡试管植株叶片的总DNA,以随机引物进行非特异性扩增,计算了遗传相似系数与遗传距离,并进行了聚类分析.从20条随机引物中挑选出3条扩增条带清晰、重复性好的引物OPC-8、OPD-8和OPE-10,共检测出22条DNA谱带,其中多态性条带18条(占82%).结果表明,9种北柴胡试管植株间均存在不同的遗传差异,根据绘制的聚类图可将9种不同类型的北柴胡聚类为4组.说明RAPD多态性分析从分子水平上一定程度反映出不同类型晋产北柴胡之间的遗传多态性.
Nine Bupleurum chinense DC test tube plants established by plant tissue culture technique were used as materials. The total DNA of the leaves of Bupleurum chinense DC was extracted by CTAB method. The random primers were used for non-specific amplification. Genetic similarity coefficient and genetic distance were analyzed and clustering analysis was carried out.From the 20 random primers, three primers OPC-8, OPD-8 and OPE-10 with clear bands and good repeatability were selected 22 DNA bands, of which 18 were polymorphic bands (82%). The results showed that there were different genetic differences among the nine kinds of Bupleurum test tube plants. According to the drawn cluster chart, nine different types Bupleurum cluster was 4 groups, indicating that RAPD polymorphism analysis at molecular level to some extent reflect the different types of Jin Buplearum genetic polymorphism.
