为探讨KAI1基因的表达与结肠癌转移的关系,用Northern blot及原位杂交分析法对10例正常人及24例结肠癌患者组织中的KAI1 mRNA表达进行检测。结果显示,在正常及结肠癌组织中均有不同程度的KAI1 mRNA表达,原发性结肠癌伴淋巴结转移的KAI1 mRNA表达水平明显低于无淋巴结转移者(P<0.01);原位杂交的KAI1 mRNA表达与Northern blot分析进行对比,发现高水平KAI1 mRNA出现在多数原发性结肠癌细胞质中,而肿瘤周围间质相对呈低表达。结论:KAI1 mRNA表达在结肠癌的抗转移中具有一定的作用。
To explore the relationship between KAI1 gene expression and colon cancer metastasis, KAI1 mRNA expression in 10 normal subjects and 24 colon cancer patients was detected by Northern blot and in situ hybridization analysis. The results showed that KAI1 mRNA expression was different in normal and colon cancer tissues. The KAI1 mRNA expression level of primary colon cancer with lymph node metastasis was significantly lower than that without lymph node metastasis (P<0.01); KAI1 in situ hybridization Comparison of mRNA expression with Northern blot analysis revealed that high levels of KAI1 mRNA were present in the majority of primary colon cancer cells, whereas the surrounding mesenchyme was relatively low. Conclusion: The expression of KAI1 mRNA plays a role in the anti-metastasis of colon cancer.
