目的探讨亚低温处理大鼠脑缺血再灌注损伤后缺血侧脑组织神经干细胞(NSC)增殖的动态变化。方法采用大鼠局灶性脑缺血再灌注损伤模型,大脑中动脉阻塞2 h,再灌注损伤3,7,11,14,18, 28 d,用免疫组织化学方法分别检测假手术组、对照组和亚低温组缺血侧脑组织5-溴脱氧尿嘧啶核苷(BrdU)阳性细胞数。结果假手术组缺血侧脑组织存在少量BrdU阳性细胞,亚低温组在缺血再灌注损伤3,7,11, 14,18,28 d时缺血侧脑组织Brdu阳性细胞数均明显多于对照组,且亚低温组缺血侧脑组织NSC的增殖高峰期较对照组延长。结论亚低温可促进大鼠脑缺血再灌注损伤后缺血侧脑组织NSC的进一步增殖。
Objective To investigate the dynamic changes of proliferation of neural stem cells (NSC) in ischemic brain tissue after mild hypothermia in rats with cerebral ischemia-reperfusion injury. Methods The model of focal cerebral ischemia-reperfusion injury in rats was established. The middle cerebral artery was blocked for 2 h and reperfusion injury was performed at 3, 7, 11, 14, 18 and 28 days. The sham operation group and control group were respectively detected by immunohistochemistry The number of BrdU positive cells in the ischemic brain tissue of mild hypothermia group and mild hypothermia group. Results There was a small amount of BrdU positive cells in ischemic brain tissue in sham operation group. The number of Brdu positive cells in ischemic brain was significantly higher in hypothermia group at 3, 7, 11, 14, 18 and 28 days after ischemia / reperfusion injury Control group, and the peak of proliferative NSC in ischemic brain tissue in hypothermia group was longer than that in control group. Conclusion Mild hypothermia can promote the proliferation of NSC in the ischemic brain tissue after cerebral ischemia-reperfusion injury in rats.
