将豌豆根瘤菌共生质粒pJB5Jl、含紫云英根瘤菌共生基因的重组质粒pRaZ15及含苜蓿根瘤菌共生基因的重组质粒pRmSL26导入紫云英根瘤菌野生型菌株7653R.然后对3种转移接合子的共生固氮效率进行了比较全面的测定。结果表明:转移接合子7653R(pJB5Jl)的共生固氮能力较出发菌株7653R大幅度提高,而且表现出较高的竞争结瘤能力,转移接合子7653R(pRmSL26)在固氮酶活性、植物干重及结瘤数方面亦显著高于对照菌株7653R;转移接合子7653R(pRaZ15)虽然在固氮酶活性及植物干重方面没有发现显著性差异,但结瘤数显著减少。
The recombinant plasmid pRaZ15 of pea Rhizobium symbiotic plasmid pJB5J1, the symbiotic gene containing Rhodotria maritima and the recombinant plasmid pRmSL26 containing the symbiotic gene of alfalfa rhizobia were introduced into the wild type strain 7653R of Rhizobium japonicum. Then we carried out a comparatively comprehensive determination of the symbiotic nitrogen fixation efficiency of the three kinds of metastasis junkers. The results showed that the symbiotic nitrogen fixation ability of the transconjugants 7653R (pJB5Jl) was significantly increased compared with the parent strain 7653R, and showed high competitive nodulation ability. The transformant 7653R (pRmSL26) The number of neoplasms was also significantly higher than that of the control strain 7653R. Although there was no significant difference in the activity of nitrogenase and the dry weight of plant 7653R (pRaZ15), the number of nodules decreased significantly.
