目的从噬菌体七肽展示库中筛选与β3整合素结合的多肽序列,并进行鉴定。方法以β3整合素为靶分子对噬菌体七肽展示库进行8轮亲和筛选,获得与β3整合素结合的噬菌体克隆,挑选结合力强的克隆进行DNA测序,推导出呈现的多肽序列,应用生物信息学软件进行多肽序列分析和同源性分析。结果经8轮亲和筛选从噬菌体展示七肽库中挑选80个克隆进行DNA序列测序及生物信息学分析,发现TGVKGPG、LPLTPLP、KLTSSPT、SPVGPLP和DHRNHLV肽段有较高的重复机率,与汉坦病毒囊膜糖蛋白氨基酸序列有较高的同源性。结论通过对噬菌体展示肽库的淘筛,获得与β3整合素结合的高亲和性多肽,为进一步设计和获取高活性的抗汉坦病毒短肽提供参考资料。
OBJECTIVE: To screen and identify the β3 integrin binding peptide sequences from phage display library. Methods The β3 integrin was used as a target molecule to screen the phage display peptide library by 8 rounds of affinity screening. The phage clones that bound with β3 integrin were obtained. The clones with strong binding ability were selected for DNA sequencing. The presented polypeptide sequences were deduced, Informatics software for polypeptide sequence analysis and homology analysis. Eighty rounds of prokaryotic expression plasmids were selected from 80 phage display peptide libraries for sequencing and bioinformatics analysis by 8 rounds of affinity screening. The results showed that TGVKGPG, LPLTPLP, KLTSSPT, SPVGPLP and DHRNHLV peptides had a high probability of repetition, Virus glycoprotein amino acid sequence has a higher homology. Conclusion By screening the phage displayed peptide library, the β3 integrin-binding high-affinity peptide is obtained, which provides a reference for further design and acquisition of highly active anti-Hantavirus short peptide.
