目的探讨促性腺激素释放激素(Gn RH)在雌二醇(E2)诱导大鼠青春期启动提前中的作用。方法 18日龄雌性SD大鼠分别经口给予玉米油和E250μg·kg-1,连续给药5 d。观察大鼠阴道开口时间及卵巢组织变化;Western蛋白印迹法检测下丘脑的Gn RH、G蛋白偶联受体54(GPR54)和磷脂酶C(PLC)蛋白表达水平。结果与玉米油对照组相比,E2组大鼠阴道开口时间显著提前约12.2 d(P<0.01)。卵巢组织切片可见黄体,且下丘脑Gn RH,GPR54和PLC蛋白表达水平分别增加47%,55%和56%(P<0.05)。结论 E2诱发的大鼠青春期启动时间提前可能与调节下丘脑Gn RH,GPR54和PLC表达相关。
Objective To investigate the role of gonadotropin-releasing hormone (Gn RH) in early adolescent initiation of estradiol (E2) -induced puberty in rats. Methods 18-day-old female Sprague-Dawley rats were orally administered corn oil and E250μg · kg-1 for 5 days. The time of vaginal opening and the changes of ovarian tissue were observed. Western blotting was used to detect the expression of Gn RH, G protein-coupled receptor 54 (GPR54) and phospholipase C (PLC) protein in the hypothalamus. Results Compared with the corn oil control group, the vaginal opening time in E2 group was significantly advanced by about 12.2 days (P <0.01). Ovarian tissue sections showed corpus luteum, and the expression of Gn RH, GPR54 and PLC in the hypothalamus increased by 47%, 55% and 56%, respectively (P <0.05). Conclusion E2-induced adolescent start-up time may be related to the regulation of hypothalamus Gn RH, GPR54 and PLC expression.
