目的:寻找一种有效的提高腺病毒的滴度而又不损害其活性的方法。方法:腺病毒原夜升化冻干(sublimation drying)后溶解稀释,通过斑点形成实验、免疫组化检验其感染活性。结果:升化冻干后稀释的腺病毒液感染滴度达到4.4××1012pfu。结论:冻干、稀释是一种提高腺病毒滴度的有效方法。
OBJECTIVE: To find an effective way to increase the titer of adenovirus without impairing its activity. Methods: The adenovirus was sublimation dried and then dissolved and diluted. The plaque formation assay and immunohistochemistry were used to test the infection activity. Results: The titer of liquid dilution of adenovirus after the increase of lyophilization reached 4.4 × 1012pfu. Conclusion: Lyophilization and dilution is an effective way to improve the titer of adenovirus.
