目的检测Krüppel样因子5(Krüppel-like factor 5,KLF5)在小鼠舌黏膜癌变模型中的表达,探讨KLF5与口腔黏膜癌变的关系。方法将42只C57BL/6J小鼠随机分为阴性对照组、4-硝基喹啉-1-氧化物(4NQO)处理组。阴性对照组不做任何处理,4NQO组以4NQO水溶液(50μg/ml)代替饮用水饲养,共16周。实验第24周末处死全部动物,取舌组织进行HE染色及KLF5平均光密度值分析。结果 HE染色显示,对照组小鼠舌背上皮为正常角化的复层鳞状上皮,饮用4NQO水溶液的小鼠舌黏膜上皮出现了不同程度的异常增生及鳞癌。平均光密度测定显示,与正常口腔黏膜上皮相比,KLF5在重度异常增生和鳞癌中的表达明显下降(P<0.01)。结论 KLF5可能在口腔黏膜上皮癌变过程中作为肿瘤抑制基因发挥重要作用。
Objective To detect the expression of Krüppel-like factor 5 (KLF5) in mouse tongue carcinogenesis model and to explore the relationship between KLF5 and carcinogenesis of oral mucosa. Methods Forty-two C57BL / 6J mice were randomly divided into a negative control group and a 4-nitroquinoline-1-oxide (4NQO) treatment group. Negative control group without any treatment, 4NQO group with 4NQO aqueous solution (50μg / ml) instead of drinking water feeding, a total of 16 weeks. At the end of the experiment, all the animals were killed at the end of the 24th week. HE staining and average optical density of KLF5 were taken from tongue tissues. Results Hematoxylin and eosin staining revealed that the lingual dorsal epithelium of mice in the control group had normal keratinized stratified squamous epithelium, and the tongue epithelial mucosa of 4NQO aqueous solution showed abnormal proliferation and squamous cell carcinoma in varying degrees. The average optical density measurement showed that the expression of KLF5 in severe dysplasia and squamous cell carcinoma was significantly lower than that in normal oral mucosa (P <0.01). Conclusion KLF5 may play an important role as a tumor suppressor gene in carcinogenesis of oral mucosa.
