目的:对在体外仿生提取牛蒡苷元的制备方法进行研究,优化筛选牛蒡苷元的酶水解条件、提取工艺等,得到最适工艺流程。方法:以酶浓度、酶解时间、底物浓度为考察因素,用Box-Behnken设计-响应面分析法对牛蒡苷元水解条件进行优化。结果:牛蒡苷元最佳制备条件:转化温度50℃,pH4.8,酶液浓度0.44 U/mL,反应时间46.81 min,底物浓度0.29 mg/mL,牛蒡苷元转化率可达到90.94%。结论:该研究为体外制备牛蒡苷元提供了一定的借鉴价值。
OBJECTIVE: To study the preparation of arctigenin in vitro and optimize the enzymatic hydrolysis conditions and the extraction process of arctigenin to obtain the optimal technological process. Methods: Enzyme concentration, enzymatic hydrolysis time and substrate concentration were used as the factors to investigate the optimization of arctigenin hydrolysis conditions using Box-Behnken design-response surface methodology. Results: The optimum conditions of arctigenin preparation were as follows: the conversion of arctigenin reached 90.94% at 50 ℃, pH4.8, enzyme concentration 0.44 U / mL, reaction time 46.81 min, substrate concentration 0.29 mg / mL. Conclusion: This study provides some reference value for the preparation of arctigenin in vitro.
