目的探讨对氧磷酶(PON)中PON155M/L和PON2148A/G基因多态性与高血压的关系。方法采用PCR-RFLP方法检测201例高血压组和94例对照组的PON155M/L和PON2148A/G基因多态性。结果与对照组比较,高血压组血浆PON、总超氧化物歧化酶(T-SOD)活性明显降低,丙二醛(MDA)浓度显著升高;PON155LM基因型、M等位基因频率,PON2148GG/AG基因型、G等位基因频率较对照组明显增多;PON155LM基因型的PON和T-SOD活性较LL基因型明显降低,而收缩血压、舒张血压显著增高;PON2148AG和GG基因型的PON活性较AA基因型者明显降低,GG基因型的MDA、AG基因型的收缩压和舒张压则显著升高。logistic回归分析显示PON155LM基因型、PON2148GG/AG基因型是高血压的危险因素。结论PON155M等位基因、PON2148G等位基因是高血压的危险因素,此等位基因携带者血浆PON活性降低。
Objective To investigate the relationship between PON155M / L and PON2148A / G polymorphisms and hypertension in paraoxonase (PON). Methods The polymorphisms of PON155M / L and PON2148A / G in 201 hypertensive patients and 94 healthy controls were detected by PCR-RFLP. Results Compared with the control group, plasma PON and total superoxide dismutase (T-SOD) activity in hypertensive group were significantly decreased and MDA concentration was significantly increased; PON155LM genotype, M allele frequency, PON2148GG / PON and T-SOD activity of PON155LM genotype was significantly lower than that of LL genotype, while systolic blood pressure and diastolic blood pressure were significantly increased; PON activity of PON2148AG and GG genotypes was significantly higher than that of control group AA genotype was significantly lower, GG genotype MDA, AG genotype systolic and diastolic blood pressure was significantly increased. Logistic regression analysis showed that PON155LM genotype and PON2148GG / AG genotype were risk factors for hypertension. Conclusion The PON155M allele and PON2148G allele are the risk factors for hypertension, and the plasma PON activity of these alleles is decreased.
